Establishment and Application of the Controllable Modified MD-300 Chemostat
Objective:The aim of this study was to modify an existent MD-300 chemostat suitable for complicated oral bacteria ecologic study. Methods:The brooder of MD-300 Chemostat was modified, in which incorporated removable and replaceable hydroxyapatite (HA) disks, and the chemostat's key parameters were regulated for primary application. Chemostat conditions of analogous oral environment were that the medium was TSB broth with 2.5g/L porcine gastric mucin, the volume 1200ml, the medium flow rate adjusted to 50ml/h, culture pH maintained at 7.0+/-0.1, temperature 37+/-0.1°æ and air was N2, CO2, H2(80:10:10). Results: Artificial plaque biofilms could conventional develop on HA dishes and bacteria adhesion to the surface of different resin deture material could be detected in the modified MD-300 chemostat. Conclusion:The study concluded that the modified MD-300 chemostat has considerable potential in studying artificial dental plaque biofilm development and bacteria ecologic relationship, as well as in comparing bacteria adhesion to different materials under analogous oral environment.Supported by grant 30070813 from National Natural Scientific Foundation.
