Association of 1607bp SNP with the susceptibility to aggressive periodontitis
Objectives:A single nucleotide polymorphism (SNP) was described in the promoter - 1607 bp of the human MMP-1 gene, and this polymorphism has been associated with risk of cancer metastasis and inflammatory diseases. In this paper, we will study the possible relationship between the MMP-1 polymorphism and aggressive periodontitis (AP). Methods:Genomic DNA was obtained from whole blood samples from 40 AP patients and 52 controls. Genotypes of MMP-1 promoter -1607bp were determined by (restriction fragment length polymorphism -polymerase chain reaction (RFLP- PCR). Results:In the controls, the 2G allele was observed with a frequency of 49%, while in the AP patients; the 2G allele was seen in 68.7%, and the distribution of 2G allele had significantly differences between AP patients and Healthy controls(p<0.05). The genotype 2G/2G was found in 52.5% of the AP, and 23.1% in the controls. Differences in genotype distribution also had significantly differences between AP patients and Healthy controls(p<0.05).Conclusions:The results show that a SNP in the MMP-1 promoter ¨C 1607 bp is associated with Chinese patients with aggressive periodontitis.
