P2X3 receptors expression in the rat trigeminal ganglion
The main process of Orthodongtics is the process of tooth movement through orthodontic force .when the tooth and periodontium is stressed ,a series of responses and change will occur ,such as the pain which is the main problem occurred during the tooth movement,the sensations are transmitted to the corresponding parts in CNS by primary afferent nerves in pulp and periodontium, the trigeminal sensory ganglion neurons is the nociceptive primary sensory neurons.The P2X3 receptor is a ligand ¨Cgated cation channel activated by the binding of extracellular adenosine 5'-triphosphate(ATP),an angent that has been suggested to have a role in the nociceptive pathway after tissue and nerve injury, Aim:The purpose of this study was to examine the precise regulation of the P2X3 receptor expression in the trigeminal ganglion sensory neurons after the nociceptive stimulation by orthodontic tooth movement force.,using immunohistochemistry°¢western blotting and in situ hybridization with an oligonucleotide probe respectively. Methods:Male Sprague-Dawley rats weighing 200-250g were used ,the mimic tooth movement appliance are used in experimental guoup rats .the animals are sacrificed after 4h,12h,1d,2d,3d,5d and 7d. Results:The P2X3 receptor and P2X3 mRNA expression is transiently upregulated and anterogradely transported in trigeminal primary sensory neurons after orthodontic tooth movement and that P2X3 receptor and P2X3 mRNA may play role in the pathomechanism of nociceptive in primary sensory neurons during orthodontic clinic treatment.
