Method: From 100 caries-free human teeth, 25 teeth were immersed in chloramine (0.5%), 25 in sodium azide (2.5%) and 25 in sodium chloride (0,9%) for at least 4 weeks. 25 teeth were processed directly after extraction without the influence of a culture medium. Standardized buccal class V cavities were prepared in all teeth, half in the enamel and half in dentin, by using water-cooling. All cavities were prepared with the etching system ‘Vococid’ by VOCO and the adhesive system Syntac ® Classic. In each group 5 teeth were filled with the following composite materials according to the manufacturer’s guidelines: Grandio ® SO, Grandio ® SO Heavy Flow, Grandio ® SO Flow, Venus Diamond Flow ® and Venus ® Diamond. By using a dye penetration test (methylene blue) we examined microscopically to compare the total margin with the defekt marginal lenth. The statistical significance was tested by multiple ANOVA and post.-hoc test by Duncan (p<0.05).
Result: With an average defect size of 9% to 82%, sodium chloride was found to be the least suitable culture medium. Regardless of the culture media, there is more penetration of methylene blue in the area of cement than the enamel. It was further revealed that Venus Diamond Flow ® achieved the best results in all culture media, with an average defect size of 9% to 75%.
Conclusion: The influence of different culture media on the bond is clearly visible and should be considered in vitro studies to maintain their validity.
