Effects of CAD/CAM all-ceramic materials on the viability of fibroblasts

Objectives: The purpose of this in vitro study was to evaluate the effects of seven CAD/CAM all-ceramic materials (VITA Mark II (VM), IPS Empress CAD (EC), IPS e.max CAD (EM), InCoris TZI (IC), VITA Suprinity (VS), Lava Ultimate (LU), Cerasmart (CS)) on the fibroblast cell viability.
Methods: CAD/ CAM blocks were cut using a low speed diamond saw and 24 disk-shaped samples (diamater=5 mm, height=2 mm) were prepared from each test material. IC, EM, and VS discs were sintered according to the manufacturer’s protocol. Medium extracts were collected at 1st, 3th and 7 th days for each group and tested using L929 cell line. Cytotoxicity was evaluated using XTT assay and apoptosis was determined by Annexin-V staining. Data were analyzed using one-way ANOVA, post hoc Bonferroni tests at a significance level of p<0.05.
Results: The cell viability results among CAD/CAM material groups after 1st, 3th and 7th days of incubation periods showed statistically significant differences (p<0.05). At all incubation periods, resin nanoceramic (LU) showed highest cell viability while leucite reinforced CAD/CAM material (IPS Empress CAD) (EC) showed lowest cell viability among all materials. Only EC material presented a significant difference at different incubation periods (p<0.05). Other materials did not present any significant difference at different incubation periods (p>0.05).
Conclusions: It was concluded that the biocompatibility of new generation CAD/CAM materials has to be improved in order to achieve a safer biologic structure.
