Effect of Isoflavone, Estrogen and Association on Bone Defects Density

The estrogen replacement therapy presents adverse effects and the phytotherapy has become reliable. Objective: Therefore, the aim of this work was to evaluate, through digital radiography, the influence of the treatment with estrogen, isoflavone isolated and associated with estrogens on bone repair of tibias of ovariectomized rats. Methods: Fifty 3-month-old rats, 40 ovariectomized and 10 rats sham-ovariectomized, were used. The treatment began 30 days after the ovariectomy, and the ovariectomized rats received water, estrogen, isoflavone and the association of isoflavone with estrogen. Twenty days after the beginning of the treatment a bone defect was created in the tibia, and the animals received the treatment until the sacrifice, which occurred 7 and 21 days after bone defect creation. Digital radiography was taken to analyze the mean density in the center of the lesion. The value was assessed by two evaluators, and the correlation intra and inter evaluator was high (r>0.85; p<0.05) to all variables. Results: After the application of ANOVA test, statistics difference was found between the ovariectomized animals and the sham group (p=0.004). Considering the treatment, the ANOVA and Tukey's test showed statistic difference in the density means (p<0.05) between the sacrifice (7 days: 56.81; 21 days: 66.47) and between the administration of isoflavone associated with estrogen (74.99) compared to estrogen (63.08), isoflavone isolated (51.03) and water (57.46). Conclusions: It was concluded that ovariectomy diminished the radiographic density and that treatment with the isoflavone associated with estrogen resulted in increase of bone repair density in ovariectomized rat tibia (Support: CAPES).
