Effect of Leukovorin Calcium (LV) and 5-Fluoracyl (5-FU) administration over the oral tissue of Wistar rats. Structural Study.

Introduction: Several workers have proved that high doses of cytostatics administration over 5 consecutive days, intensify toxicity in oral tissue of experimental animals, namely macroscopic changes such as facial swelling, stomatitis, enlargement of salivary glands as well as histopathologic disorders. The purpose of this report was to analyse structural Submandibular Gland (GSM) and tongues of Wistar rats LV+ 5FU treated by correlating cytostatics dose administration with side effects on these tissues.
Material and Methods:20 Wistar rats,fed ad libitum and exposed to a 12 hr. light- dark cycle were used. Subjects were divided into 2 groups: 1- Control (C;n:8) and 2- LV +5-Fu;n:12 treated rats. LV was i.p. administered (10 mg/kg BW) +5-Fu (50 mg/kg BW) for 5 consecutive days. On the seventh day of chemotherapy, SMG and tongues were dissected .
Results: SMG was affected in mucosa acines, with vacuolated cytoplasm, disarray and stroma disrupture. Serose acines were preserved. The excretory duct revealed discontinuous epithelium with more flattened and picnotic basal cells. Significant vasodilation in connective tissue with plenty of perivascular mastocytes and plasmocyte infiltrate. In tongue: decrease in corneal stratum, perivascular mastocytes and vasodilation . No bacterial infection was found.
Conclusion: these disorders would account for effects such as stomatitis, hyposalivation and dysgeusia observed during oncologic treatment.
