BiodentineTM: Leachate Analysis In Artificial Saliva With Differing pH.

Objectives: Biodentine™ is a calcium-silicate based material which is advocated for many clinical applications including as a temporary restorative material. Tricalcium-silicate cements produce calcium hydroxide as a byproduct of hydration. To date, there is limited research into the physical properties which may support Biodentine’s™ use as a restorative material. Furthermore, there are no published studies investigating the leachate profile of Biodentine™ in differing pH solutions. The aim of this research is to to evaluate the calcium and silicon ion release of Biodentine™ exposed to artificial saliva solutions with differing pH.
Methods: Biodentine™ (Septodont, Saint Maur des Fosses, France) samples were mixed according to manufacturer’s instructions and condensed into 2ml RNA extraction tubes (9mm internal diameter) open only at one end. They were allowed to set for 12 minutes and weighed; after which, the samples (n=224) were randomly immersed in 15ml eppendorf tube containing 10ml of artificial saliva modified to a pH of either 4.4 (lactic acid/sodium lactate buffer), 7.2 (HEPES buffer) or 10.4 (glycine/NaOH buffer). Eight samples were removed from each pH solution at each time period of 10, 30, 60, 180, 1440, 4320 and 10080 minutes respectively. The solutions were assessed for calcium and silicon concentration using inductively coupled plasma atomic emission spectroscopy (ICP-OES).
Results: Following the initial pilot phase, it was determined that in artificial saliva of pH=7.2, calcium ion concentration increased over time, while the increase in silicon ion concentration was not significant.
Conclusions: There is no relationship between the release of calcium and silicon ions (solution pH=7.2) from Biodentine™ over time. Furthermore, the effect of pH and increased time periods on the calcium and silicon ion release from Biodentine™ in artificial saliva will be determined.
