EDTA-Etching and Plant-Based Polyphenols Boost Resin-Dentin Bonds Durability

Objectives: We have recently demonstrated that treatment of EDTA-demineralized dentin with plant-based polyphenols (PB-P) increases mechanical and biochemical stability of this tooth substrate. This study investigated whether treatment with these PB-P [luteolin (LT), galangin (GL), and proanthocyanidin (PAC)] affect the bond strength of an etch-and-rinse adhesive to EDTA-etched dentin.
Methods: Thirty-two non-carious extracted human third molars were collected and had the occlusal surface removed using a low-speed diamond saw under water-cooling to expose a flat dentin surface and further polished through 600-grit paper. Samples were randomized to be etched with phosphoric acid ((PA),15s) or 17%EDTA ((EDTA), 60s), and subsequently treated with one of the following primer solutions: 1) ultrapure water (W, -control); 2) 0.2% LT; 3) 0.4% GL, or 4) 1% PAC (+control). Primers were scrubbed on PA-etched or EDTA-etched dentin surfaces during 30s. Any excess of primer was removed with lab wipe. Two layers of adhesive ExciTE-F (Ivoclar, Armhest, NY, USA) were applied over primed surfaces for 30s, air-dried for 5s and light-cured for 10s (VALO, Ultradent Products, South Jordan, UT; irradiance>1200mw/cm2). Composite buildups (Empress-Direct, Ivoclar) were made with four 1.5-mm increments and polymerized for 20s. Samples (n=4 teeth/group) were stored in ultrapure water (37oC/24hr), sectioned into beams for microtensile testing (µTBS) and tested immediately (IM-µTBS) or after 9-month water storage (9mon-µTBS) using a universal testing machine (EZ-LX, Shimadzu, Columbia, MD, USA). Data were analyzed by ANOVA (post-hoc Tukey, α=0.05).
Results: For PA-etched specimens, no statistically significant differences in IM-μTBS were detected among groups (p>0.05). However, reduction in 9mon-μTBS was significantly higher for W (-control) in comparison with PB-P-treated samples (p<0.05). For EDTA-etched specimens no statistically significant differences were detected in either IM- or 9mon-μTBS among groups (p>0.05) (Table).
Conclusions: Combination of EDTA-etching and treatment with PB-P played a synergistic effect on durability of resin-dentin bonds.
