Fluoride, Calcium and Phosphate Recharge by an RMGI Protective Material

Objectives: Ion release decreases over time; recharge by topical treatments or saliva might prolong the benefits. Determine fluoride recharge of an RMGI by 5000 and 950 ppmF toothpastes, SnF2 rinse, and fluoride varnish, and calcium and phosphate recharge by Artificial Saliva.
Methods: Materials shown in Table 1. Cured 20x1mm VXT disks (n=5) were stored @37°C in 25ml deionized water(DIW). At 1hr,6hr,24hr,2d,3d,4d,7d leachate was collected/replaced. Groups A-C: C5K or CTC toothpaste, or PM rinse treatments were applied at 7d,8d,9d; aliquots were collected immediately before treatment, and 1,2,4,6hr after each treatment. Toothpaste treatment was immersion 2min in 25ml DIW:toothpaste=3:1; PM treatment was immersion 1min in 25ml 7:1= DIW:PM. Group D: varnish was applied at 7d, removed with razor scraping and acetone wipe @8d; aliquots were collected 1,2,4,6,24,48,72,96hr after DIW re-immersion. Group E: at 7d disks were immersed in 25ml of Artificial Saliva(Newby,2006); @8d the disks were DIW rinsed, immersed in 25ml DIW; aliquots were collected at 1,2,4,6,24,48,72,96hr after re-immersion. Fluoride was measured with ion-specific-electrode after adding TISAB-II; calcium and phosphorus concentration were measured via ICP-AES. Data after treatment were compared to immediately before treatment via one-way paired T-test (p<0.05).
Results: Tables 2-4 show ion release rate. Within each treatment and recharge period, groups with superscript * are pre-recharge(baseline), H are higher than pre-recharge, S are statistically not different from pre-recharge.
Conclusions: VXT showed increased fluoride release up to 6hr after recharge by NaF toothpastes or SnF2 rinse, and up to 4d by NaF varnish. VXT showed increased Ca release up to 4hr and P release at least 24hr after recharge by Artificial Saliva. VXT can recharge and re-release F, Ca, and P ions by a variety of sources; this may aid in providing an environment conducive to protection and remineralization of tooth structure.
