GALNT14-rs9679162 Genotype Predicts Postoperative Outcome in Oral Squamous Cell Carcinoma

Objectives: Objective
To comprehensively examine the relationship between OSCC and GALNT14 genotype, the research objectives have been delineated into the three facets :
1.1. Objective 1 : Exploration of the distribution of GALNT14-rs9679162 genotypes among OSCC patients
1.2. Objective 2:Clarify the connection of GALNT14-rs9679162 genotypes and risk factors among OSCC patients
1.3. Objective 3:Find out the relation of GALNT14-rs9679162 genotypes and survivorship rate
Methods: 2.1. Subjects
A total of 113 cases of OSCC were acquired from the Changhua Christian Hospital Tissue Bank.
2.2. DNA extraction
DNA was extracted employing a TRI Reagent RNA isolation kit.
2.3. Genotyping
PCR was conducted to obtain GALNT14-rs9679162, employing the specified primers: Forward: 5′-TCACGAGGCCAACATTCTAG-3′, Reverse: 5′-TTAGATTCTGCATGGCTCAC-3′, Genotyping was performed under the following conditions : 95 °C for 1 min, 58 °C for 1 min, and 72 °C for 1 min, for a total of 40 cycles. The 172 bp PCR products were isolated from the gel and sequenced utilizing a 377 DNA sequencer, strictly following the manufacturer′s instructions.
Results: Results
3.1. GALNT14-rs9679162 Genotypes Frequency in OSCC
Among the 113 OSCC cases, 29 cases were GG type (26%), 48 cases were TG type (42%), and 36 cases were TT type (32%).
3.2. The Connection between GALNT14-rs9679162 Genotypes and Risk Factors among OSCC
Among OSCC patients who engaged in alcohol consumption, betel nut chewing, and cigarette smoking, the incidence of the non-TT genotype (GT and GG genotype) was higher.
3.3. The relation of GALNT14-rs9679162 Genotypes and survivorship rate
Among OSCC patients undergoing chemotherapy, the survival rate for those with the GALNT14-rs9679162 GG genotype was notably lower.
Conclusions: Conclusions
The GALNT14-rs9679162 genotypes can serve as an indicator of the therapeutic outcome in OSCC patients.
