Antibacterial Activity of Tea Tree Oil Nanocapsules Coated With Chitosan Against Important Bacteria for Oral Infections

Objectives: This work aimed to develop poly(ξ-caprolactone) nanocapsules carrying Tea Tree Oil (TTO) coated with chitosan (NP-TTO-CH), with satisfactory physicochemical characteristics and antimicrobial activity against oral bacteria.
Methods: The suspensions containing nanocapsules were prepared by the interfacial precipitation method of the preformed polymer. Then, they were coated with 2% chitosan and characterized according to their mean hydrodynamic diameter (Z-average), pH, polydispersity index (Pdi) and zeta potential (Zp). The essential oil encapsulation efficiency was also evaluated by means of gas chromatography. For the biological characterization, tests of minimum inhibitory concentration and minimum bactericidal concentration were performed.
Results: The Z-average of the NP-TTO-CH was 282.5 ± 2.5 nm, the mean Pdi was 0.264 ± 0.004, the mean Zp was +39.4 ± 0.5 mV and the mean pH was 4.85 ± 0.53. The average encapsulation efficiency of the TTO was 89.89 ± 1.29%. The NP-TTO-CH showed microbiostatic antimicrobial activity against the bacteria Staphylococcus aureus ATCC BAA977, Staphylococcus aureus ATCC 33591, Staphylococcus aureus ATCC 25905, Enterococcus faecalis ATCC 51299, Enterococcus faecium ATCC 6569, Streptococcus mutans ATCC 25175, at a concentration of 5mg/ml and against Streptococcus sobrinus ATCC 27351 and Klebsiella pneumoniae ATCC 2874 at a concentration of 10mg/ml, during the first 48 hours of the experiment. This activity was maintained at 72 hours of experiment against the bacteria S. aureus ATCC 33591, S. aureus ATCC 25905, E. faecalis ATCC 51299 and S. mutans ATCC 25175 at a concentration of 10mg/ml.
Conclusions: Nanocapsules of Tea Tree Oil coated with 2% chitosan proved to be promising against the microorganisms tested, and may become treatment alternatives in several areas of Dentistry.
