Biocompatibility of Endodontic Medications and Materials on Dental Pulp Mesenchymal Stem Cells

Objectives: Background: Dental pulp mesenchymal stem cells (DPMSCs) are responsible for regenerating the dentin-pulp complex following tissue injury/disease. Thus, the medications and materials employed in endodontics treatment must be biocompatible with DPMSCs.
Objective: To assess the effect of commonly employed endodontic medications and materials on DPMSCs
Methods: DPMSCs were isolated from sound third molars extracted due to impaction in healthy adults (17-18 years old) and characterized (-ve for CD34, CD45, HLA-DR, and +ve for CD73, CD90, CD 105). Following characterization, the DPMSCs were exposed to several endodontic medications and materials including calcium hydroxide, doxycycline, potassium iodide, triamcinolone, and glutaraldehyde. The dose and time-dependent effects of these endodontic medications and materials on DPMSCs viability (MTT assay) were assessed. In addition, an annexin V assay was performed to assess the effect on the DPMSC apoptotic status.
Results: DPMSC vitality was attenuated in both a time and dose-dependent fashion by all the tested medications and materials, while the apoptotic status of the DPMSCs was significantly augmented following exposure to doxycycline and glutaraldehyde. Calcium hydroxide, potassium iodide, and triamcinolone did not induce any significant apoptosis in DPMSCs.
Conclusions: Though all the tested endodontic materials and medications attenuated DPMSCs vitality, only doxycycline and glutaraldehyde induced significant cell death and could be considered to be relatively less biocompatible than calcium hydroxide, potassium iodide, and triamcinolone.
