Glass Ionomer Doped with Copper-silver Nanoparticle on Cariogenic Dual-species Biofilm

Objectives: This study aimed at evaluating the effect of a glass ionomer cement (GIC) incorporated with copper-silver nanoparticle (CuNp) on dual-species biofilm formation of Streptococcus mutans (SM) and Candida albicans (CA).
Methods: The CuNp was incorporated into GIC (Gold Label 9, GC Corp) at two different concentrations: 0.05% (w/w) and 0.1% (w/w). Discs of CIC without CuNp were used as control. Dual-species biofilms were formed on the surface of the discs in 24-well plates contained TSBYE culture medium supplemented with 1% sucrose, for 72 hours. The biofilms were then collected and processed. Colony-forming units (CFU) of SM and CA were recovered on Mitis salivarius-bacitracin and Sabouraud dextrose agar, respectively. Three disks of each group were subjected to biofilm formation per experiment and the microbiological experiment was done in triplicate (n=9). The antimicrobial effect was determined by CFU, dry weight and confocal laser scanning microscopy (CLSM).
Results: Data analyses (α=0.05) showed no statistically significant differences among the tested groups in relation to either dry weight mean (p=0.405) or CA CFU counts (p=0.126). One-way ANOVA (p=0.008) and Dunnett multiple comparisons test showed reduced SM CFU mean counts for GIC doped with CuNp at 0.05% (6.86×109±6.97×109) and at 0.1% (6.91×109±4.09×109) when compared to the control GIC (1.03×1010±9.19×1010). Qualitative CLSM analyses corroborate with CFU results.
Conclusions: CuNp-doped-GIC showed limited activity on dual-species biofilm formation when compared to conventional GIC. Further studies on the effect of this incorporation must be tested.
