Effect of Phytosphingosine on Staining Resistance and Microhardness of Enamel

Objectives: The purpose of this study was to evaluate the protective effect of phytosphingosine (PHS) on dental enamel with regard to staining in vitro.
Methods: Ninety-six specimens of bovine teeth (6 x 6 x 2 mm) were prepared, and initial color (Easyshade, VITA), microhardness (HMV-2, Shimadzu) and fluorescence (Matlabs software, Matworks) measurements were performed. Specimens were randomly separated into four groups according to the treatments to which they were submitted: Distilled water (control); Human saliva (HS); Phytosphingosine (PHS); PHS + HS. After that, specimens (n = 6) were submitted to staining procedures: Distilled water (immersion for 30 days - control); Coffee (15 minutes, twice a day, for 15 days); Black tea (as described for Coffe) and cigarette smoke (20 cigarettes/sample). Then, final color, microhardness, and fluorescence measurements were performed. Color and fluorescence data were analyzed using 2-way ANOVA test, Tukey, p<.05; while microhardness was analyzed by Kruskal-Wallis test, p<.05, finding the lowest values in groups stained with coffee.
Results: It was found that PHS treatment reduced staining of the enamel by cigarette smoke, both in the presence and absence of a salivary pellicle.
Conclusions: PHS did not alter the color of the salivary pellicle. In line with previous research, PHS prevented tissue loss in the presence and absence of a salivary pellicle, as could be deduced from changes in microhardness measurements.
