Type IX Collagen Distribution In Mouse Periodontal Ligament
Objectives: The periodontal ligament (PDL) has long been known for its ability to resist tensile forces in the same manner as tendons. More recently, the PDL has been shown to also resist compressive loads in a manner similar to cartilage. Type IX collagen is a member of the FACIT group of collagens, and is normally covalently bound to type II collagen in cartilage and other tissues that contain type II collagen. Previous studies have shown the "cartilage-specific" molecule, type IX collagen, to be present in the human PDL. Interestingly, type IX has also been shown to be expressed in the healing alveolar bone after tooth extraction. The purpose and function of collagen IX in the PDL is still unknown, especially since there is no type II collagen to which it can covalently bond. The objective of this study was to determine whether or not collagen type IX is present in the mouse PDL. Methods: Mandibles from Gli1-CreERT2;Rosa26tdTomato C57/B6 mice s were fixed in 4% paraformaldehyde, decalcified, embedded in Tissue-Tek® and sectioned. Immunostaining was performed using a commercially available α(1)IX polyclonal antibody and a FITC labeled secondary antibody. The sections were treated with DAPI, and the fluorescent images captured on a confocal microscope. Results: Diffuse immunostaining above background was noted in the PDL. There also appeared to be staining above background in isolated areas of the alveolar bone. Conclusions: Collagen type IX is present and generally distributed throughout the mouse periodontal ligament. There also appears to be small amounts of positive signal in the alveolar bone.
