Host Defense Peptides Associated to MTA in the Dentin-Pulp Complex Reparative Process

Objectives: Dental carie or complex dental fracture may lead to the dental pulp exposure and the direct pulp capping treatment allows the remaining pulp tissue preservation. Currently, MTA is the most suitable material for such treatment, despite its disadvantages and questionable antibacterial action. Otherwise, due to their antimicrobial and immunomodulatory properties, host defense peptides (HDPs) can act as adjuvants in repair processes. Thus, this work aims to evaluate the HDPs (IDR1018 and DJK-6) potential alone or associated with MTA in dental pulp cells, for dentin-pulp complex repair.
Methods: This project was approved by the UCB Human Research Ethics Committee (2.209.368 / 2017). Antibacterial assays were initially done with micro-dilution growth inhibition and biofilm eradication assays. Furthermore, primary pulp cells were cultivated, and cell viability and migration were evaluated. Cell morphology was also analyzed using scanning electron microscopy (SEM).
Results: Results demonstrated that DJK-6, IDR1018 and MTA were uncapable of inhibit planktonic bacteria, but clearly presented antibiofilm properties. Peptides and MTA extract were non cytotoxic to pulp cell culture. Furthermore, the DJK-6 and IDR1018 combination with MTA improved cell migration induction, when compared to MTA alone (p<0.05, by one-way ANOVA test). There was also evidence of HDPs and MTA aggregation on the cell surface, detected by SEM analysis.
Conclusions: Thus, it was observed that DJK-6 and IDR1018 can improve MTA properties, emerging as possible therapeutic agents for pulp capping.
