Dental and Periodontal Phenotype in Kalirin Knockout Mice
Objectives: Kalirin, a novel GTP-exchange factor protein found in skeletal tissue in osteoclasts and osteoblasts and KALRN global deletion leads to low bone mass in mice. This led us to question whether Kalirin deletion might cause significant changes in dental and periodontal phenotype of mice. We analyzed the maxillae and mandible of Kalirin knockout (Kal-KO) and wild type (WT) mice to determine the effects of Kalirin on dental and periodontal structures using micro-computed tomography (µCT) imaging. Methods: Analyses were performed on maxillary 1st molar tooth and its surrounding structures. Mean and standard deviations for tooth volume, crown volume, root volume and periodontal ligament (PDL) volume were calculated. In addition, a single slice from the central region of the first mandible molar was analyzed for total bone volume (excluding the molar and incisor) and lingual cementum-enamel to alveolar bone crest (CEJ-AC) distance. This distance is roughly equivalent to the clinically assessed periodontal pocket, which is believed the nidus of inflammation. Statistical significance between Kal-KO and WT control mice was determined by standard Student T-test and one-way ANOVA with Fisher’s LSD post hoc tests when appropriate. A 5% significance level was used for all tests. Results: Kalirin KO mice had significantly decreased crown volume as well as 10% increase in CEJ-AC distance compared to wild-type mice (P<0.05). However, no significant differences were found for the tooth, root and PDL volumes and percentage bone area. Conclusions: Our results suggest that Kalirin deletion may have an effect at the crown formation of the tooth as well as increase alveolar crest bone resorption in adult mice.
