PHTALOX® Antimicrobial Action and Cytotoxicity: in vitro Study

Objectives: To compare the inhibitory effect on Streptococcus mutans and cytotoxicity of PHTALOX®, a new phythalocyanine, and sodium fluoride.
Methods: The in vitro inhibitory activity of a 01% aqueous solution of PHTALOX® (PHOX) and 1450 ppm of sodium fluoride (NaF) against planktonic cells of S. mutans (ATCC 25175) were determined. A 108 cells/ml bacterial suspension were exposed to PHOX , NaF and PHOX + NaF for 1, 3 and 5 min under agitation. The number of cells in the remaining suspension was determined by plating method and expressed in CFU/mL. Furthermore, the cytotoxicity of both substances was evaluated. Standardized inoculum containing 6x105 Vero cells in DMEM were added onto 96-well plate, followed by incubation (37°C, 24 hours, 5% CO2). Sequentially, 200 µL of DMEM previously exposed for 24 hours to PHOX (0,1%) or containing 1450 ppm NaF was added. After that, 100 μL of MTT (0,5 mg/mL) was added followed of further incubation for 4 hours. Subsequently, 100 µL of DMSO was added and OD values were determined a spectrophotometrically (570 nm). The assays were made in triplicate and statistical analyzed by ANOVA and Tukey's test (p <0.05).


Results: Statistically significant decrease was observed after treatment with PHOX (all exposure periods), NaF (5 minutes) and PHOX + NaF (3 and 5 minutes) when compared to control PHOX was significantly more effective in S. mutans inhibition than NaF after 3 and 5 min of exposure. Exposure to PHOX alone was more effective than the combination with NaF after 1 and 5 min of exposure. Moderate cytotoxicity (about 50% of viability) and severe cytotoxicity (less than 30% of viability) were observed for PHOX and NaF, respectively.


Conclusions: PHTALOX® showed better results in S. mutans inhibition and lower cytotoxicity compared to sodium fluoride, and may be a promising alternative in oral care.
