Antibiofilm Action of PHTALOX®-containing Oral Care Formulations

Objectives: To verify the antimicrobial action of a new commercial phthalocyanine (PHTALOX®) incorporated in oral care formulation with and without sodium fluoride against biofilms of S. mutans.
Methods: The groups studied were: Dentifrice with PHTALOX® 0,1% + sodium fluoride 1450 ppm (DPHOX); Colgate Total 12 New formula - sodium fluoride 1450 ppm (CT12); Oral gel with PHTALOX® 1,0% (GPHOX); Oral rinse with PHTALOX® 0,1% (ORPHOX) Positive control: sterile water (ST); Negative control: (CLX) - chlorhexidine digluconate 0,12%. The 24 hours biofilm of Streptococcus mutans (ATCC 25175) were formed at 37°C and 5% of CO2 onto 96-well plates with TSB broth (1% sucrose). Sequentially, the biofilms were treated on room temperature during 5, 10 and 30 minutes under shaking. After that, the biomasses were determined by violet crystal technique (VC) and cell viability was performed by MTT assay. The values of both were determined thought OD values under 600 nm (VC) and 570 nm (MTT) using a microplate reader. The tests were made three times in triplicate and statistical analyzes were determined using ANOVA, Tukey’s and paired t-test (p <0.05).
Results: The decrease in biomass and cell viability of biofilms at all treatments had been observed when compared with positive control. Furthermore, the biomass after treatment with DPHOX and CT12 was significantly lower compared to the other treatments in all studied treatment times, although there was no difference between them. The biofilms treated 5 minutes with DPHOX and ORPHOX have shown statistical decrease of cell viability when compared with CT12. Moreover, have been observed cell viability decrease when compered the ORPHOX with CLX in 5 minutes. There was not relevant difference in other treatment times.
Conclusions: The study revealed an antimicrobial potential of PHTALOX® incorporated in oral care formulations, associated or not with sodium fluoride, against S. mutans biofilm.
