Novel Dentifrices Exhibit Enhanced Effects Toward Achieving Good Oral Health
Objectives: To determine the remineralization, biofilm regrowth, abrasivity, oxidation of salivary biomolecules of two stabilized chlorine dioxide dentifrices and the USP Reference. Methods: New CloSYS® Silver toothpaste contains sodium fluoride (0.21%), stabilized chlorine dioxide, and sodium myristoyl sarcosinate.CloSYS® Anticavity toothpaste contains sodium fluoride (0.24%), stabilized chlorine dioxide, and sodium lauroyl sarcosinate. Remineralization was investigated by treating lesions on bovine teeth enamel with toothpaste slurry followed by cyclic remineralization and acid challenge treatment. Fluoride uptake and Surface Micro Hardness (SMH) were determined after 20 days of treatment. Biofilm regrowth was studied by brushing the biofilm grown on bovine enamel and observing the extent of regrowth of residual biofilm by confocal microscopy. RDA values were determined using standard methodologies. Oxidation of salivary biomolecules was monitored by 1H NMR spectroscopy. Results: Enamel fluoride uptake by Anticavity and Silver toothpastes was 96.1% and 27.4% higher than the USP Reference, respectively. SMH of Anticavity and Silver toothpastes was 37%, and 78% higher than the USP Reference, respectively. Silver toothpaste exhibited higher remineralization although fluoride uptake was lower than Anticavity toothpaste. Silver toothpaste inhibited the polymicrobial mixture of biofilm vs. control 6 h after the biofilm was brushed, demonstrating significant superior bactericidal action (p = 0.0388) within the biofilm. RDA score of Silver toothpaste (142) was lower than the Anticavity toothpaste (207), both toothpastes exhibiting RDA below 250 recommended by ANSI/ADA. Anticavity toothpaste effectively oxidized salivary biomolecules such as L-pyruvate and L-methionine in 30 seconds. Conclusions: The higher remineralization, lower RDA, reduced regrowth of polymicrobial biofilm, and faster rate of oxidation of salivary biomolecules of the CloSYS® dentifrices is attributed to the symbiotic effect of sodium fluoride, stabilized chlorine dioxide, and respective sarcosinate. CloSYS® Silver toothpaste exhibits unsurpassed remineralization, lower RDA, and reduced biofilm regrowth compared to Anticavity toothpaste and the USP Reference.
