A Novel nanosized Bioglass Enhance MG-63 Cells Proliferation and Gene Expression

Objective: To investigate the effects of a novel nanosized 58S bioglass (nano-58S) on proliferation and specific osteoblast markers expression in MG-63 cells.

Method: MG 63 cells were cultured in 1% w/v nano-58S and 58S particulates conditioned medium and DMEM respectively. Proliferation was assayed on day 1,3,5,7,10 by MTT. Gene expression of alkaline phosphatase (ALP), Cbfa 1, collagen type I and osteocalcin was observed on day 2,4,7 by real-time RT-PCR. At day 7, 14 and 21, ALP expression and matrix mineralizaion was assayed with BCIP/NBT ALP Color Development Kit and Alizarin red S solution.

Result: Nano-58S group showed significantly enhanced cell proliferation and expression of ALP, Cbfa 1 and OCN  in a time-depenednt manner compared with the control group. The expression of Cbfa 1 on day 4 and OCN on day 7 was significantly higher than that in 58S group. There was significantly more ALP protein expression and matrix mineralization in nano-58S BG group than 58S group.

Conclusion: The novel nano-58S bioglass enhanced proliferation and osteogenic markers gene expression in MG 63 cells. Compared with 58S bioglass, nano-58S showed a higher activation of Cbfa 1 and OCN gene expression.
