Modulatory effect of low dose beta2-blocker without affecting heart function

Objective: Some animal studies have suggested that bone remodeling is under beta-adrenergic control. Although pharmacological studies on adrenergic receptors have provided controversial results. Thus, the objective of the present study was to analyze the effects propanolol given in different doses in a rat model of periodontal disease induced by ligature. Methods: Rats were assigned to one of the following groups: 1) animals without ligature receiving empty vehicle; 2) ligated-animals receiving empty vehicle; 3) ligated-animals receiving 0.1 mg of propanolol; 4) ligated-animals receiving 5mg of propanolol and 6) ligated-animals receiving 20 mg of propanolol. Thirty days after induction of periodontal disease, the animals were anesthetized and heart functions were assessed by catheterization of the left ventricle. Gingival tissues were removed and assessed for IL-1β quantification by ELISA and ICAM-1 and RANKL expression by western blot analysis. Results: We observed that the low dose of propanolol significantly reduces the IL-1β expression in gingival tissue. Also, 0.1 and 5 mg of propanolol inhibited the expression of ICAM-1 and RANKL. Important no significant effects on heart hemodynamic parameters were found in rats receiving the lowest dose of propanolol on the other hand, medium and high doses had a negative effect on heart functions. Conclusion: These results suggest that low doses of β-blockers may have a therapeutic utility in the treatment of periodontal disease. Financial support: CNPq 471305/2009-0 and FAPEMIG 097/09
