Cytotoxic Effects of Orthodontic Band Adhesives

Objectives: The aim of the present study was to evaluate the cytotoxic effects of three different resin modified orthodontic band adhesives (RM-OBA). Methods: RM-OBA; Bisco Ortho Band Paste LCTM (Bisco), Multi-Cure Glass Ionomer Band CementTM (3M Unitek ) and Transbond Plus Light Cure Band AdhesiveTM (3M Unitek) were prepared and the samples were extracted in 3ml of BME with 10% new born calf serum for 24h. The L929 cells were plated (25.000cells/ml) in wells of 96 well-dishes, and maintained in a humidified incubator for 24 hours at 37{degree sign}C, 5%CO2 and 95% of air. After 24h incubation of the cells, the incubation medium was replaced by the immersed medium in which the samples were stored. Then L929 cells were incubated in contact with eluates for 24h. The cell mitochondrial activity was evaluated by the methyltetrazolium test (MTT). 12 well used for each specimen and MTT tests applied two times. The data were submitted to the statistically analyzed by One -way ANOVA and Tukey HSD tests. Results: Results with L-929 fibroblasts demonstrated that all freshly prepared RM-OBA materials reduce vital cell numbers (p>0.05) in comparison to control group. Our data demonstrate that all materials showed significant cytotoxicity compare to control group. Conclusion: Results indicate that tested orthodontic bonding composites are not suitable for clinical application, and further studies used different test methods are needed for all RM-OBA.
