Candida albicans biofilm development characteristics on different Poly(methyl-methacrilate)

Objective: the aim of this study was to evaluate the characteristics of biofilm development of Candida albicans on two different Poly(methyl-methacrilate) resins (PMMA). Methods: Two different Candida species (Candida albicans ATCC 90028 and Candida albicans SC5314) were allowed to develop biofilm in two PMMA with different polymerization method: Lucitone (LC) and Acron (AC). Different time points (adhesion, 24, 48 and 72 hours) were compared. Surface roughness (SR) and surface free energy (SFE) of PMMA specimens were previously measured. Biofilm activity were measured by XTT. Confocal images were used to measure biomass, average thickness, biofilm roughness, diffusion and the of LIVE/DEAD cells proportion in the different biofilm development stages. Cell counts were used to estimate the number of cells in those biofilms. Results: LC and AC showed no difference regards SR and SFE means (p>0.05). Biofilm bioactivity showed different results for each specie (p<0.001). Cells counts and confocal images showed a biomass increased according the different time points evaluated. No difference regarding the substrata was found. Conclusion: It can be conclude that the different polymerization method of PMMA did not interfered with biofilm development and the difference detected were related to the intrinsic characteristic of each Candida specie.
