Comparison of HA coated and oxidized fixture using PRP grafting

Objective: Recent platelet rich plasma (PRP) grafting materials can be applied osteoplasty. In this study, we examined four wall bone defect models that initially could not be corrected. An implant containing a mixture of PRP and autogenous bone was then applied to examine whether or not the bone defect could corrected using implant. Materials and Methods: Six female beagles of weight 10-12 kg were used. Under general anesthesia, the right and left premolars were extracted and after bone healing the peripheral alveolar bone was removed to make a bone defect model. Autologous PRP and bone collected from the mandible were ground up and mixed to create the PRP grafting materials. Using this material, an implant was then placed in the bone defect sites. An oxidized surface fixture was placed on the mesial side, and an HA coating fixture was placed on the distal site. The surface was sutured to complete the operation. Animals were injected with calcein and euthanized 2, 4 and 8 weeks after the operation and the mandible was removed. The alveolar bone of the premolar was removed and the condition of the bone was observed using light microscopy and confocal laser scanning microscopy. No obvious ossification was observed 2 weeks after the operation, but advanced ossification was noted after 4 and 8 weeks. Results: Our results suggest that the PRP grafting material promoted bone formation and fixed the implant, even under condition of no bone fixation. In addition, a fixture with an HA coating surface promoted bone formation inside threads to a much greater extent than an oxidized surface fixture. Conclusions: The PRP graft could be useful for early wound healing and could be effective as adjuncts in gnathic bone regeneration and fixture integration.
