Distribution of Human Lactoferrin on the Periodontally Affected Dental Root

Objective: Human lactoferrin (hLf) is an iron-binding protein found in milk, tears and neutrophils. A recent study suggested that hLf inhibits lipopolysaccharide (LPS) interaction with CD14. LPS from periodontopathic bacteria permeates dental root surfaces exposed to periodontal pockets, eliciting an inflammatory response. Thus, hLf released from migrating neutrophils may be involved in the inflammatory response. The objective of the present study was to investigate the distribution of hLf on dental roots. Methods: Teeth not salvageable by periodontic treatment were extracted with the informed consent of patients. Powdery samples were prepared from extracted roots with a round bur-equipped electric motor-driven system. The samples were collected and partitioned into three groups on the basis of location: Layers I, II and III (from the root surface to the pulp cavity). Specimens were eluted in PBS. Following centrifugation, the supernatant was used to measure hLf levels. Subsequently, extracted teeth were incubated with anti-hLf antibody and Alexa 532-conjugated secondary antibody. Fluorescence measurements were performed on a Molecular Imager FX Pro¨. In addition, hLf was introduced to J774.1 murine macrophage-like cells infected with A. actinomycetemcomitans in order to evaluate the effects on apoptotis. Results: Significantly higher hLf levels were detected in Layer I from the area with calculus in comparison to Layer I from the area without calculus. hLf levels decreased from Layer I to Layer III. The area displaying fluorescence was in agreement with the area with calculus. LDH activity, DNA fragmentation and TNF-a production in the infected J774.1 cells were reduced upon addition of hLf. Conclusion: These findings suggested that hLf released by neutrophils is bound to LPS leading to inhibition of the immunobiological effects of LPS to host cells, which results in host protection.
