Antibacterial Effects of Brazilin and Hematein from Caesalpinia sappan
Objective: New agents are needed to treat methicillin-resistant Staphylococcus aureus (MRSA) from acute oral infection. In the present study, we isolated antibacterial components from Caesalpinia sappan (C. sappan) by bio-assay guided fractionation, and they were identified as brazilin and hematein by MS, 1H-NMR, and 13C-NMR. Methods: C. sappan was extracted with 80% MeOH, the extract was concentrated in vacuo, and the residue partitioned between H2O and CHCl3, EtOAc, and n-BuOH, respectively. The fractions were assayed for growth inhibitory activity against MRSA. Column chromatography of the most active fraction on silica gel, Sephadex LH20, and prep-HPLC gave purified compounds. The structure of compounds as identified by MS, 1H-NMR, and 13C-NMR. Results: The EtOAc fraction from solvent partition possessed greater antibacterial pontential than other fractions. The MICs of the fraction obtained from column chromatograpy by silica gel and Sephadex LH20 against MRSA 33591 were 0.625 mg/ml. The fraction was separated by prep-HPLC to purified compounds. The compounds were identified as brazilin and hematein by MS, 1H-NMR and 13C-NMR. The MICs of brazilin and hematein against MRSA 33591 were 0.156 mg/ml. Conclusion: These results suggest that brazilin and hematein isolated from C. sappan by bioactivity-guided fractionation may have antibacterial activity against MRSA.
