Methods: Affymetrix gene chip was utilized to determine differential gene expression between human papillomavirus infected, immortalized human oral keratinocytes (HOK16E6E7) and normal human oral keratinocytes (NHOK). Northern blot analysis was performed to examine p55CDC expression in various immortalized and cancer cell lines.
Results: Affymetrix microarray analysis demonstrated that p55CDC was over-expressed in HOK16E6E7 by 16 fold over NHOK. Compared to NHOK, the expression of p55CDC was increased in both immortalized HOK16E6E7 and HOK16B that harbored the entire HIV-1 genome. HaCaT, a spontaneously immortalized epidermal keratinocyte also contained . P55CDC expression was further expressed in other head and neck cancer cell lines, including Hep-2, SCC-4, SCC-9, Cal27, Cal33, HaCaT, Hela and HOK16B-BaP-T. High level expression of p55CDC was also observed in cancer cell lines from different tissues, including several highly proliferative leukemia cell lines, such as K-562, MOLT-4 and HL-60.
Conclusions: These data suggested that p55CDC gene maybe detected in preneoplasia and serve as early diagnosis and of head and neck cancer.
