3D-Printed PCL-Gelatin Membrane For Guided Tissue Regeneration
Objectives: The use of guided tissue regeneration (GTR) has significantly increased in periodontology. In the present study, 3D membranes of polycaprolactone (PCL)-gelatin with various porosities were fabricated as a dual functioning scaffold for potential GTR application. Methods: The PCL membranes were fabricated using 3D-printing after optimizing the parameters. The membranes were then aminolyzed with 1,6-hexanediamine and post-treated via glutaraldehyde (2.5 %wt). Next, the membranes were washed repeatedly with copious amount of deionized water and PBS. Finally, the membranes were immersed into a porcine gelatin (10 %wt) to attach the gelatin molecules to PCL membranes. The attachment of gelatin and aminolyzed PCL was confirmed by FTIR spectra. The morphology of the scaffolds was evaluated using 3D-laser microscope and scanning electron microscope (SEM). Biocompatibility of the scaffolds was measured using different cell types of dental pulp stem cells, osteoblast and fibroblast. The effect of scaffolds on osteogenesis was analyzed using alkaline phosphatase activity. Results: The FTIR results confirmed aminolysis of 3D-printed PCL membrane and successful attachment between gelatin molecules and aminolyzed PCL. The morphology characterization revealed that a thin layer of gelatin with a thickness of 250-500 µm uniformly encompassed the 3D-printed aminolyzed PCL membrane. The in vitro results showed that the fabricated GTR has no significant cytotoxicity on the dental pulp stem cells, osteoblast, and fibroblast. Moreover, the results demonstrated that the scaffolds can affect osteogenic differentiation of dental pulp stem cells by modifying ALP activity. Conclusions: In this study, the PCL-gelatin scaffolds showed that it can be used for osteogenic differentiation and suggested a new approach in improving osteogenic GTR.
