Planktonic Metabolic Activity Using Mixed Species Salivary Inoculum

Objectives: Evaluate the anti-bacterial efficacy of Dual zinc plus Arginine containing dentifrice by utilizing in-vitro planktonic salivary inoculum model compared to negative and positive control dentifrices.
Methods: The anti-bacterial performance of dentifrice is determined here by using planktonic salivary Short Interval Kill Time assay (SIKT). The SIKT assay is used to measure efficacy of formulas post 30 second treatments. The viability of bacteria is quantified using traditional plate count method (CFUs). Results are reported as log reductions from the untreated control.
Results: Planktonic salivary SIKT shows Dual zinc plus Arginine to have better efficacy compared to the competitive stannous fluoride variants. Dual zinc plus Arginine had a log reduction of 4.93 compared to the log reductions of the competitive low water and high water stannous fluoride variants, 3.77 and 3.60 log reductions respectively. In addition, the Dual zinc plus Arginine also shows statistically significant differences from the negative control with a log reduction of 0.57.
Conclusions: Based on the early assessment for the Dual zinc plus Arginine dentifrice efficacy using SIKT, it shows that there is a greater effect in reducing bacterial viability when compared to competitive stannous fluoride dentifrice variants. The early antibacterial efficacy allows for fewer bacteria to remain in the mouth, suggesting less biofilm formation over the course of time. The results provide evidence that the dual zinc plus arginine formula is very effective at targeting salivary bacteria.
