Angiogenic Protein Release After Photobiomodulation by Stem Cells

Objectives: This study aimed to evaluate the protein express after Low-level Laser Therapy (LLLT) with two different energy densities on stem cell from human exfoliated deciduous teeth (SHED).
Methods: SHED were cultured in MEMα 10% FBS (Fetal Bovine Serum) between the 5th and 8th passage. The groups were divided according to density of energy: G1(2,5J/cm2;10 mW, 10 s); G2 (3,7J/cm2;15 mW, 10 s); and Control (not irradiated). Aluminium-gallium-indium-phosphide (InGaAlP) diode laser (Twin Flex Evolution MMOptics® – São Carlos, Brazil) was used the 660nm wavelength (red); tip diameter of 0.225 cm; output beam area of 0.04 cm2 laser; continuous mode and applied in single time. After 6h, 12h, and 24h from irradiation, the protein expression was acquired through ELISA Multiplex assay. Data were submitted to statistical analysis by ANOVA 2 criteria, followed by Tukey test.
Results: Over time in the supernatant, all groups presented an increase of the levels of VEGF-C, VEGF-A, and PLGF, while only the irradiated groups had a VEGFR2-expression peak at 12h. In the lysate, the control group and G2 presented decreased of VEGF-A, PECAM-1, and PLGF, while control group and G3 showed a decrease of VEGF-C, VEGF-A, and PDGF. The groups irradiated had different peaks of protein expression, G2 had higher expression of VEGF-C and PDGF at 6h, and G3 had a higher expression at 12h of proteins PECAM-1, FGF-2, and PLGF.
Conclusions: Photobiomodulation produced stimulatory and inhibitory effects on angiogenic protein synthesis by stem cells.
