Antifungal Effect of The Phyllanthus Amarus Partitions Against Candida
Objectives: Phyllantus amarus is a medicinal plant popularly known as “chancapiedra”. The aim of this study was to evaluate the antifungal effect of the butanolic, chloroformic and aqueous partition of Phyllantus amarus against Candida albicans, C. tropicalis, C. krusei and C. parapsilopsis. Methods: The Phyllantus amarus crude extract was completely solubilized with methanol (MeOH) and chloroform (CHCl3), water (H2O) was added, followed by the solubilized crude extract. Small portions of CHCl3 were added to the column containing the sample under study, all the concentrated chloroformic phases were called chloroform residue. In another column butanol (BuOH) was added with the Phyllantus amarus sample, the butanolic phases was concentrated until obtaining a solid which was called BuOH residue. The aqueous phase was removed and frozen for subsequent lyophilization, which was called aqueous residue. These three partitions (chloroformic, butanolic and aqueous) were diluted in dimethylsulfoxide DMSO (20mg/ml, 40 mg/ml). The antifungal effect of the three plants and their combinations were evaluated by well diffusion against Candida albicans, C. tropicalis, C. krusei and C. parapsilopsis and an inhibitory halo reading was performed using a vernier. The controls were DMSO and Clorhexidine 0.125%. Results: Chloroformic residue had no effect against any Candida obtaining halos of 0 mm. Butanolic residue had positive halos of inhibition against Candida albicans: 20mg/ml: 13.04±0.51mm,40mg/ml: 15.04±0.79mm; Candida tropicalis: 20mg/ml: 14.05±1.63mm,40mg/ml: 15.92±2.13mm; Candida krusei: 20mg/ml: 14.17±1.64mm,40mg/ml: 17.08±1.51mm; Candida parapsilopsis: 20mg/ml: 14.12±1.70mm,40mg/ml: 16±1.15mm. Aqueous residue had positive halos of inhibition against Candida albicans: 20mg/ml: 12.82±1.52mm,40mg/ml: 13.5±1.92mm; Candida tropicalis: 20mg/ml: 14.21±0.84mm,40mg/ml: 16.68±1.71mm; Candida krusei: 20mg/ml: 16.31±3.11mm,40mg/ml: 15.8±2.31mm; Candida parapsilopsis: 20mg/ml: 14.66±1.83mm,40mg/ml: 16.81±1.15mm. Clorhexidine 0.125%: Candida albicans: 18.32±4.6mm, Candida tropicalis: 21.35±4.24mm, Candida krusei: 25.16±2.51mm, Candida parapsilopsis: 24.2±3.66mm. Conclusions: Butanolic and Aqueous partitions of Phyllantus amarus have antifungal effect against Candida albicans, C. tropicalis, C. krusei and C. parapsilopsis.
