The Effect of CGF on the Regeneration of Injured Peripheral Nerves
Objectives: Peripheral nerve injury is one of the complications after oral implant or tooth extraction. Several studies have shown that previous product of concentrated growth factor(CGF)——platelet rich plasma(PRP)can increase the number of local regenerated nerves and the thickness of myelin, and CGF was found to increase the sciatic functional index. However, the mechanisms remain unclear. Therefore, this study aims to investigate the influence and mechanism of CGF on the injured peripheral nerve regeneration. Methods: In vitro, Schwann cells(SCs)from SD rats were obtained and then treated with different concentrate CGF conditional medium. CCK8 and transwell migration assay were used to examine the effect of CGF on cell proliferation and migration. Real-time PCR was performed to evaluate the expression of neurotrophic factors. Western Blot was employed to explore the expression change of ERK1/2 phosphorylation in the process. In vivo, rats were divided into sham (n=8), control(n=8) and CGF group(n=8). We observed and analyzed the regeneration of crush injured inferior alveolar nerve. Results: In vitro, we noted an obvious proliferation in the 50% concentrate medium, which presumably by activating ERK pathways. Migration assay showed that CGF promoted the migration of SCs. However, PCR showed that CGF had no promotion effect on the secretion of neurotrophic factors. In vivo, we found that CGF group, compared to control group, promoted the regeneration of injured inferior alveolar nerve. Conclusions: Our findings demonstrate that CGF promotes peripheral nerve regeneration,ERK1/2 phosphorylation might play a crucial role in this process.The results suggest that the injured nerves around implants are expected to be repaired.
