Effect of 3D-printing-scaffolds Based on PLA in Culture With hDPSC
Objectives: To print scaffolds based on PLA and evaluate their effects in culture with hDPSC (Human Dental Pulp Stem Cells). Methods: The scaffolds were designed, printed and characterized with atomic force microscopy (AFM) and stereomicroscopy. The enzymatic and hydrolytic degradation of the scaffold was determined by UV-visible. The hDPSC were isolated, cultivated and characterized under international guidelines. Cytotoxicity and cell adhesion was determined by the MTT assay and immunofluorescence. The experiments were performed in triplicate of three independent experiments (n= 12) and the data were analyzed with Shapiro-Wilk normality and t-student tests, a conventional culture dish was used as a control group. Results: The scaffolds have an average roughness of 10 μm and fill porosities of 95% are observed. Enzymatic and hydrolytic degradation was significantly reduced (p<0.05) at 166 hours of agitation (6%). The hDPSC showed fibroblastoid morphology, adherence to plate, favorable differentiation to chondrogenic, adipogenic and osteogenic lineage, positive markers to CD105 +, CD90 +, CD73 + and negatives to CD14-, CD34-, CD45-. The cytotoxicity of the scaffolds slightly reduces the number of viable cells (85±8%), while the cell-scaffold interaction significantly increases (p<0.05) cell proliferation (125±5%). Conclusions: The 3D print scaffolds based on PLA-conventional shows favorable effects in the interaction with hDPSC which can be an excellent option as a biomaterial for regenerative dentistry.
