Osteoblast Differentiation Induced by a Prototype S-PRG Filler-containing Endodontic Sealer
Objectives: Surface reaction type Pre-Reacted Glassionomer (S-PRG) filler shows bioactive and antimicrobial properties due to its ability to releases multiple ions including borate, fluoride, silicate and strontium. The aim of this study was to evaluate the effects of extracts from a prototype S-PRG filler-containing endodontic sealer on osteoblast differentiation. Methods: The S-PRG filler-containing sealer (S-PRG; Shofu, Japan) and a silica filler-containing experimental sealer (SF; control; Shofu) were mixed according to the manufacturer's instructions, and inserted into plastic cylinders (8 mm diameter, 3 mm height). Following complete setting, the specimens were placed in 10 ml of culture media for 24h with shaking, and filtered media were used as sample extracts. A clonal osteoblast cell line, Kusa-A1 cells, was cultured in the presence of the sample extracts. Cell proliferation and osteoblastic differentiation were evaluated with WST-1 assay and RT-PCR, respectively. Nodule formation was induced by culturing the cells in an osteogenic medium containing ascorbic acid and beta-glycerophosphate. Phosphorylation of Erk was analyzed with the western blot. NPS 2143 hydrochloride was used as an antagonist of calcium-sensing receptor (CaSR). Results: Cell proliferation was slightly down-regulated in the presence of S-PRG extracts. mRNA expression of Alkaline phosphate and Bone sialoprotein, typical osteoblastic markers, was up-regulated by S-PRG extracts, which was blocked by NPS 2143. Nodule formation was promoted in the presence of S-PRG extracts. Phosphorylation of Erk was enhanced in the presence of S-PRG extracts. Conclusions: Extracts of the S-PRG filler-containing sealer induced osteoblastic differentiation of Kusa-A1 cells, which may be mediated by Erk signaling via CaSR.
