Comparative Evaluation of Remineralization Potential of CPP-ACP and CaSP

Objectives: This study aimed to compare the Remineralization Potential of Casein phosphopeptide amorphous calcium phosphate (CPP–ACP) and Calcium Sucrose Phosphate (CaSP) on demineralized human enamel. using DIAGNOdent, contact profilometry and eSEM on demineralized human enamel in vitro.

Methods: 90 sound caries-free premolars extracted for orthodontic reasons with no detectable caries, hypoplastic lesions or white spot lesions, and a DIAGNOdent score less than 7 were selected. A 4x4mm window was created on the labial surface of selected samples. They were then demineralized (using solution with pH 4.5 containing 2.2-mM CaCl2.2H2O, 2.2-mM NaH2PO4.7H2O, 0.05-M lactic acid) for 48 hours, remineralized (using artificial saliva(control/group1), CPP-ACP (Group 2) or CaSP (Group 3)) for 7 days with 4 min topical application daily and stored in artificial saliva after washing with deionized water. Samples were evaluated at baseline, after demineralization and after remineralization. Statistical analysis was done using Students paired ‘t’ test and one-way ANOVA with post hoc analysis using tukey’s correction for DIAGNOdent and profilometric scores.


Results: As per both DIAGNOdent and profilometer testing, all three groups showed significant remineralization (p value < 0.05), however, CaSP (p vaue < 0.05) proved to be significantly better than the other two groups. As per the post hoc test for multiple comparisons, CaSP > CPP-ACP > Control.

Conclusions: Within the limitations of this in vitro study, we can conclude that both remineralizing agents have potential for remineralization of early carious lesion. However, CaSP showed better remineralization potential than CPP ACP and artificial saliva alone.
