Adhesive-assissted Biominerlization

Objectives: This study was to evaluate the adhesive-assissted intrafibrillar biominerlizations.
Methods: Polyaspartic acid (PAsp) stabilized amorphous calcium phosphate (ACP) nano-particles(PAsp-ACP)were synthesized by mixing the same volumes (10ml) of 12mM Na2HPO4 and 20mM CaCl2/480ug/ml PAsp solutions and stirred for 10 min. Two one-bottle mild self-etch adhesives (Clearfil S3 Bond, Kurraray; Adper Easy one, 3M ESPE) were used to test the remineralization ability after they were mixed with or without 25wt% PAsp-ACP and light-cured as mineralization films and adhesive films. The grids coated with single layer reconstituted type I collagen fibrils were upside down floated on artificial saliva in which mineralization films were kept under bottoms for 1, 4, 7, 14, 21 or 28 days, then they were analyzed by TEM and SAED. The cytotoxicity of PAsp-ACP particles was assessed by CCK-8 assay. Calcium and phosphate ion release from mineralization films were also evaluated in sodium chloride solution (133mmol/L, pH 7.4) for 1, 4, 7, 14, 21, or 28 days.
Results: PAsp-ACP particles had no cystoxicity. The calcium and phosphate ions released from the two mineralization films were increased during the period of 28 days. Collagen intra- and extra-fibrils were detected to be mineralized after incubation in artificial saliva containing mineralzation films since 4d.
Conclusions: Adhesive-assissted biomineraliazation exhibited a remineralization ability of type I collagen intra- and extra-fibrils and a succesive release of Ca and P ions.
