Systemic Evaluation of Bisphosphonate and Parathyroid Hormone in Rats
Objectives: This study aimed to evaluate the systemic effects in liver and kidneys in model of rats treated with local application of biphosphonate (BFs) and parathyroid hormone (PTH).
Methods: Fifteen male rats were divided into 03 groups: Group C (control), Group B (local application of bisphosphonate, 1mg/kg) and Group P (local application of parathyroid hormone solution, 20mg/dose). It was created a bone defect in the frontoparietal region and bone particles were removed and grafted back into the region. Local applications were made in the region of the defect, once a week, until the time of euthanasia. All animals were euthanized at day 60. The kidneys and livers were removed for histological analysis.
Results: It was observed liver and kidneys with normal appearance in Group C. Group B showed a beginning of a degenerative process in the liver, represented by hepatic steaosis, and in the kidneys no changes were observed. In Group P liver and Kidneys were also found with normal appearance, despite the use of medication. The histologic features are similar to those found in the control group.
Conclusions: It was concluded that the group that used PTH didn’t showed histological changes in n the kidneys and liver, as expected, and the group that used bisphosphonate presented hepatic steaosis, compatible with an initial degenerative process.
