Genotoxicity, Cytotoxicity and Phytochemical Analysis of Bromelain on Oral Cancer
Objectives: This study aims at performing a preliminary phytochemical analysis of bromelain and to check for genotoxic and cytotoxic potential of bromelian on oral cancer cell line. Methods: Preliminary phytochemical analysis of bromelain was done ,MTT assay for evaluating the cytotoxicity potential of the extract on oral cancer cell lines was performed. The genotoxicity is analysed by DNA Fragmentation. DNA isolation and DNA fragmentation is done in a step wise manner. Agarose gel electrophoresis is done and DNA is viewed using ethidium bromide under UV light. Results: Bromelain was proved to contain anti-cancerous properties.Phytoconstituents such as flavonoids, cowmarins and phenols were present in the bromelain. When genotoxicity was analysed, DNA fragmentation occurred in all the three concentrations. Bromelain also exhibited increased cytotoxicity with increased concentration. Conclusions: The proteolytic enzymes in Bromelain used as a phtyomedical compound demonstrates anti-inflammatory, anti-edematous, absorption of antibiotic drugs, anti-thrombotic, inhibition of tumor cells proliferation and exhibition of strong immunogenicity. The phyto constituents found in bromelain reveals the antioxidant property. The study exposed the cytotoxic potential and antitumor properties of bromelain. The potential to develop bromelain as an anticancer drug is a thrust area for future research in drug designing industry.
