PVM/MA Copolymer Enhances Antibacterial Efficacy of IPMP in Model Toothpaste

Objectives: To evaluate the effect of PVM/MA copolymer on the in-vitro antibacterial efficacy and solubility of 4-isopropyl-3-methylphenol (IPMP) in model toothpastes.
Methods: Pellicle coated, hydroxyapatite (HAP) discs were treated with model toothpaste formulations containing IPMP, with, and without, the PVM/MA copolymer (2% w/w). The treated HAP discs were then rinsed well to remove any adhering toothpaste formulation and further challenged by shaking for one hour in artificial saliva at 37°C. The HAP discs were then placed on top of THYE agar plates inoculated with S. mutans and incubated at 37°C for 48 hours. The distances of the inhibitory zones around the HAP discs were measured at the end of the incubation time.
Results: The discs treated with model toothpastes containing IPMP and PVM/MA copolymer showed larger zones of inhibitions on the agar plates compared to the control toothpastes containing IPMP without the copolymer. The addition of 2% PVM/MA copolymer improved the solubility of IPMP in these model toothpastes, resulting in clear solutions.
Conclusions: The addition of 2% PVM/MA copolymer to a model toothpaste formula containing IPMP improves the antibacterial efficacy of treated HAP discs compared to the same formula without copolymer. In addition, the PVM/MA copolymer improved the solubility of IPMP in this formulation.
