Cytotoxicity Evaluation of Newly Developed Thermoplastic Elastomer Root Canal Filling Material by MTT Assay

Objectives: To compare cytotoxicity of three root canal filling materials;Thermoplastic elastomer(TPE), Resilon® and Guttapercha on mouse fibroblasts cell line L-929 viability.
Methods: TPE, Resilon® and Gutta percha were incubated in MEM cell culture medium for specific periods of time and the extracted solutions were subsequently introduced to mouse fibroblasts cell line L-929. Viability of L-929 cells was evaluated using MTT assay according to ISO standard 10993-5, 2009.
Results: In TPE groups, averaged cell viabilities of 101%, 100%, 97% and 92% were observed for 1, 3, 5 and 7 days of incubation time, whereas 47-60% and 12-19% were observed for Resilon® and Gutta percha at the same time period. No morphological change of the L-929 cells in TPE groupswas observed at any exposure time. The changes were however observed partially in Resilon®groups and significantly in Gutta percha groups.
Conclusions: TPE had better biocompatibility than Resilon®and Gutta percha.
