Analysis of Calcium Dosage and Blood Count After Local Application of PTH in Rat Calvaria
Objectives: The aim of this study was to assess whether the local application of a single PTH dose changes the serum total calcium and blood count values in rats. Methods: Sixteen male rats were divided into two groups: control group (C) and PTH Group (Case). A 5 mm critical-size defect was created in the calvaria of all animals. In Group C, the defects were filled only with blood clot, whereas in the Case Group, defects were filled with a hydrolyzed collagen sponge soaked in PTH and covered with bovine cortical membrane. The blood collection, via cardiac puncture, was performed before surgery (initial) and prior to euthanasia at 15 and 60 days after surgery (final). Results: Data were submitted to descriptive and statistical analysis using the computer program Statistical Package For Social Science - (SPSS, version 20.0; SPSS Inc. Chicago, IL, USA) with 95% confidence interval. As a result, descriptively, in the PTH group, inflammation was significantly greater when compared to group C after 15 days. The final calcium dosage in Group C at the time of 60 days was significantly lower than the initial measurement of calcium. The only group which the final calcium content was higher than the initial dosage was in the 60 days PTH group, but there was no statistical difference. Conclusions: It can be concluded that the local use of PTH altered calcium dosage at the time of 60 days after surgery, suggesting that local application leads to its systemic action.
