The Effect of Cyclodextrin Complexation on Bupivacaine Cytotoxicity in Human Fibroblasts
Objectives: To evaluate the effect of bupivacaine (BUPI) alone or complexed with 2-hydroxypropyl-β-cyclodextrin (BUPI-HPβCD) on release of some pro-inflammatory cytokines and on cell-viability of human fibroblasts (FGH). Methods: Cells were grown in DMEM medium supplemented with 10% heat-inactivated FBS and 50 µg/mL gentamicin. FGH were exposed to 1 µM, 10 µM and 100 µM of bupivacaine formulations during 6 h. Cell viability were determined by XTT (in %) and live/dead assays. Cytokines (IL-1alpha, IL-6, IL-8, and TNF-alpha) quantification (ng/mL) was performed by ELISA. Kruskal-Wallis and SNK tests were used to compare formulations (alpha=5%). Results: Table 1 shows the results (median and interquartile deviation). None statistically significant differences were observed among groups regarding IL-1alpha. BUPI, independently of concentration, changed the levels of the other cytokines. BUPI-HPβCD also altered the cytokines levels in comparison to both control and BUPI groups. Both BUPI-HPβCD 1 µM and 10 µM reduced cell viability. Conclusions: The complexation with HPβCD changed the effect of BUPI on FGH cytokines release. This effect was not dependent on BUPI concentration. HPβCD also increased BUPI toxicity on FGH.
