Cytotoxicity of a Polyguanidine Compound on Dental Pulp Stem Cells
Objectives: Potent anti-tumor efficacy of CatDex (polyguanidine) has previously been demonstrated. Preliminary results show that CatDex also has antimicrobial properties demonstrated in two common oral bacteria. Its efficacy is at the same level as Chlorhexidine (CHX), the most common oral antiseptic. However, CHX damage gingival fibroblasts and an alternative oral antiseptic without this side effect would be desirable. The present study investigates the effect of CatDex on dental pulp stem cells (DPSCs). Methods: CatDex was prepared as described previously. DPSCs were isolated from third molars of young human adults by magnetic separation technique in which CD105 magnetic microbeads were used. The cells were incubated in dulbecco’s minimum eagle’s medium (DMEM-F12) with bovine fetal serum 10% and antibiotics for 24 hrs. CHX 0.12 mM (0.12%) was used as control. Incubation time was until 1 hr. The cytotoxic effect of CatDex 0.12 mM was determined by the FMCA method. Cell viability was measured at 495 nm and the percentage of viable cells was calculated. Results: The DPSCs viability was less affected by Catdex than with CHX. CatDex: after 5 min 76% viable cells, 15 min 48%, 30 min 49% and at 1 hr 41% viable cells. Corresponding figures for CHX: after 5 min 29% viable cells, 15 min 23%, 30 min 15%, and after 1 hr 18% viable cells. Conclusions: CatDex show significantly less cytotoxicity to DPSCs compared to CHX. The preliminary results indicate that CatDex could be an alternative oral antiseptic and further studies are in progress.
