Antibacterial Activity of Amphipterygium Adstringens against Streptococcus Mutans Biofilm
Objectives: This in vitro study evaluated the antibacterial activity and the chemical composition of the extract of Amphipterygium adstringens, plant used in Mexican traditional medicine, against both, planktonic and biofilm of Streptococcus mutans, the principal pathogen related with dental caries, and is commonly found in human dental plaque or biofilm. Methods: S. mutans UA159 was cultivated with brain heart infusion broth and susceptibility assay for the extract was performed according the protocol of Clinical and Laboratory Standard Institute. Broth microdilution method was used to determined MIC and colony counting (CFU/ml) on agar plates for MBC at concentrations ranging from 0.001 mg/mL to 1.0 mg/mL. S. mutans biofilm was formed on a polystyrene 6-well plate using BHI broth supplemented with 2% (w/v) of sucrose. The biofilm was treated with extract in various concentrations and inoculated on agar plate for enumeration of viable S. mutans by counting colony forming units (CFU/mL). Results: The results showed that the crude extract possess antimicrobial activity against planktonic cells at low concentration: 0.125 mg/mL for MIC and 0.31 mg/mL for MBC. The inhibition of growth of the extract on biofilm was of 1.1 mg/mL of MIC and 17.6 mg/mL for MBC. The extract of A. adstringens was submitted to chemical analyses to identify the bioactive compounds using chromatography. The compounds identified were masticadienonic acid, 3α-hydroxymasticadienonic acid, 3-epi-oleanolic, as well as β-sitosterol and phenols. Conclusions: In conclusion, the extract of A. adstringens possess a good in vitro antimicrobial activity at low concentrations against planktonic and biofilm of S. mutans, and may be a potential candidate for prevention of dental caries.
