Adhesion Inhibition of Mannan-coated Denture Resin Against Candida Biofilm
Objectives: It has been indicated that adhesion of Candida albicans and Candida glabrata to denture surfaces are related to diseases such as denture stomatitis and aspiration pneumonia. It has been known that Mannan, a Candida cell surface polysaccharide, contributes to the adhesion of the cell to mucosa and plastic surfaces. The purpose of this study is to examine the adhesion inhibition effect of Mannan-coated denture acrylic surfaces against C. albicans and C. glabrata.
Methods: After coating the denture base resin surface with Mannan and then with filtered human saliva, pre-cultured C. albicans (ATCC18804) or C. glabrata (ATCC90030) were used to form a biofilm on the surface. The amount of Candida biofilm was measured by CFU assay using washing out method with SCDLP medium for the yeast type, and by measuring the fluorescence using REDOX Indicator added to RPMI 1640 culture medium for the pseudohyphal type obtained data were analyzed using Mann-Whiteney U-test at significance level (α=0.05).
Results: Mannan had inhibitory effect against Candida biofilm adhesion in a concentration dependent manner. There was a significant difference between the 0.1mg/ml concentration of Mannan and the control (p<0.05). Furthermore, the adhesion inhibition effect of Mannan against C. albicans was confirmed both in yeast and pseudohyphal type, and continued for three days from inoculation.
Conclusions: Mannan coat on acrylic resin was effective to prevent Candida biofilm adhesion. It can be used as an effective means to maintain denture hygiene in a senior citizen society.
