Fluoridating Efficiency of MFP Containing Toothpaste in Fluoride Uptake Models

Objectives: Enamel fluoridation is one of the key mechanisms involved in caries prevention, and ability to fluoridate demineralized enamel is used to predict product’s anticaries efficacy. FDA 40 and in-vitro pH cycling are key methods used in the assessment of fluoridating potential of fluoride toothpaste. The purpose of this study was to compare enamel fluoridation of three Sodium Monofluorophosphate (MFP) toothpastes in single vs. multiple treatment models.
Methods: Demineralized human enamel specimens were prepared according to White (Caries Res.1987). A pH cycling protocol of Faller, et al (Caries Res. 1991) and a standard FDA 40 protocol were used for enamel fluoridation assessment. In FDA 40 test, a single 30 minute exposure to 1:3 slurry was carried out followed by water rinse. In pH cycling test a 4-day cycling regime was used: dentifrice treatments were interspersed with saliva and a demin challenge each day. Crest Cavity Protection (1100 F, NaF) is used as a positive control in pH cycling model. Following the treatment procedures, microdrill biopsy was used to determine fluoride uptake of each specimen.

Conclusions: Multiple treatments in a cycling model significantly enhance fluoride uptake into enamel relative to single exposure. Colgate Maximum Protection plus Sugar Acid Neutralizer (1450 ppm F, MFP) delivers enamel fluoride comparable to Colgate Cavity Protection (1100 ppm F, MFP) in both models.
