Interactive Effects of Vitamin-D3 and Soy on Oral Cancer Proliferation

Objectives: Recent studies have found soy isoflavones demonstrating inhibitory properties against many cancers. Other research has demonstrated similar effects induced by VitaminD3 (VitD). Preliminary work by this group has demonstrated interactive effects that suggest each compound may potentiate the effects of the other, amplifying their anti-tumor effects. The objective of this study was to investigate the expression of the Vitamin-D receptor (VDR) in response to VitD and soy protein extract (SPE) administration, singly and in combination, in oral cancer and normal cell lines in vitro.
Methods: RNA was isolated from oral squamous cell carcinoma cell lines (CAL27, SCC25, SCC15) and the normal oral cell line (HGF-1) following VitD (125 nmol) and SPE (10 uM) administration, concentrations approximating the normal physiologic range. Quantitative RT-PCR determined changes in VDR mRNA expression over time.
Results: VitD appeared to increase mRNA expression of VDR in CAL27, SCC25, SCC15 and HGF-1 cell lines (1.91-, 1.88, 1.95- and 1.97-fold, respectively). SPE was sufficient to increase mRNA expression of VDR in these cells (2.03-, 1.72-, 1.93-, and 1.65-fold, respectively). VitD and SPE concomitantly induced an additive effect on mRNA expression of VDR (2.81-, 2.30-, 2.18-, and 2.11-fold, respectively). Increases were inversely associated with oral cancer proliferation. Inverse effects were not observed in the normal cell line.
Conclusions: Administration of VitD and SPE induce an increase in mRNA transcription of the VDR receptor and may function in a positive-feedback loop. These pathways appear to have anti-growth effects in the cancerous cells, not observed in the normal cell line control. This may suggest research into the activation of VDR pathways that may provide alternative mechanisms that could be utilized to control oral cancer growth without significant deleterious effects on normal cells and tissues.
