Characterization and Bioactivity of Nanostructures Fabricated on Ti-6Al-4V Alloy

Objectives: A titanium nanosheet (TNS) forms on titanium in 10M NaOH at atmospheric pressure. We aimed to determine the temperatures of alkaline treatment and subsequent heat treatment to prepare nanostructures on Ti-6Al-4V titanium alloy.
Methods: Specimens were immersed in 10M NaOH at 30, 40, 50, or 60°C for 24h. Surfaces were characterized by scanning electron microscopy (SEM). Following alkaline treatment at 30°C, the specimens were heated at 200, 400, 600 or 800°C for 1h. Surface characterizations of the specimens were performed by SEM, atomic force microscopy, X-ray photoelectron spectroscopy, and X-ray diffraction. The nanostructure bioactivity was investigated by soaking in simulated body fluid (SBF) and cell adhesion of bone marrow mesenchymal stem cells.
Results: The optimal temperature of the alkaline solution was 30°C, whereas surface cracks developed on samples treated at 40, 50, and 60°C. Nanopore structures on TNS free of vanadium and aluminum induced by 10M NaOH at 30°C consolidated gradually with subsequent heat treatment at 200–600°C. However, the nanostructure was destroyed, and aluminum and vanadium oxide were formed at 800°C. Hydroxyapatite precipitated on the entire TNS surface after immersion in SBF for 14 days. Subsequent heat treatment significantly promoted cell attachment on the modified surfaces. These effects were most pronounced by heat treatment at 600°C.
Conclusions: TNS nanopore structures on Ti-6Al-4V display high mechanical stability, excellent apatite-inducing ability, and biocompatibility. This bioactive titanium alloy fabricated with 10M NaOH at 30°C and then heat treatment at 600°C may be a promising metallic material for dental implants.
